رویکرد درمانی جدید برای MS و و بیماری های مشابه
نوعی مولکول پروتئینی به نام «TSPO » وجود دارد که می توان از آن به عنوان هدف جدیدی برای یافتن روشهای درمان MSبهره جست.
17می، 2013 ساینس دیلی- آزمایش دانشمندان روی مدلهای موش مولتیپل اسکلروزیس MS)) هدف مولکولی جدیدی برای طراحی داروهایی بوجود می آورد که میتواند ایمن تر و موثرتر از داروهای تایید شده توسط FDA که در حال حاضر برای MS شناسایی شده اند باشد.
یافته های این مطالعه تحقیقی،در مجله EMBO منتشر شده و نشان می دهد مولکولی پروتئینی سبب درمان برای MS ، فلج مغزی و اختلات ژنتیکی نادر موسوم به leukodystrophies، و همه اختلالات مرتبط با از دست دادن ماده سفید بافت مغز میباشد، این ماده حامل اطلاعات بین سلول های عصبی در مغز و نخاع است.
مولکولِ هدف، پروتئین ترانسلوکاتور تحت عنوان TSPO میتوکندریایی است. این مولکول قبلا شناسایی شده بود ولی ارتباط آن با MS معلوم نبود.MS نوعی بیماری خود ایمنی است که بر اثر تخریب غلاف میلین ایجاد می شود و انتقال امواج عصبی را مختل می‌کند.
محققان اثرات درمانی TSPO میتوکندریایی را پیدا کرده اند. آنها دریافتنددر موش های مدل MS علائم بیماری بعد از درمان با دارویی به نام اِتی فوکسین که با مولکول مذکور عمل میکند، بهبود یافت.زمانی که اِتی فوکسین در موش های MS قبل از بروزعلایم بیماری مصرف شد شدت بیماری در مقایسه با موش های گروه گواه که این دارو را دریافت نکرند کاهش یافت. و هنگام اوج شدت بیماری، دارو توانست علایم MS را از بین ببرد.
به گفته «ونبین دنگ» رئیس گروه این تحقیق اِتی فوکسین اثر محاظتی در مقابل فقدان میلین دارد. این رشته ها کار انتقال پیام را از سلول های مغزی به عهده دارند.
در حال حاضر درمان قطعی برای این بیماری شناخته نشده وشیوه های درمان موجود فقط در تغییر سرعت روند بیماری مؤثر هستند.
اختلال بینایی، تاری دید، دو بینی، حرکات غیر ارادی و سریع چشم و به ندرت از دست دادن کامل بینایی، اختلالات تعادل، لرزش، عدم تعادل در راه رفتن ، سرگیجه ، ضعف وگزگز اندام ها، برخی از علائم این بیماری محسوب می شوند.
کشف این محققان نشان می دهد که TSPO میتوکندریایی می تواند به ترمیم غلاف میلین در بیماران MS و حتی به ترمیم انتقال پیام ها در سیستم عصبی مرکزی منجرشود.
به عقیده محققان درمانهای بهتری برای MS و دیگر بیماری های ناشی ازتخریب غلاف میلین لازم است اما درمان های موجود آسیب ناشی از حملات سیستم ایمنی روی غلاف میلین را ترمیم نمیکنند. این تحقیق نویدگر تحقیقات بیشتر برا ی کاربردهای درمانی TSPOمیتوکندریایی در دستیابی به داروهای سالم تر و ایمن تر برای MS و دیگر بیماری های خود ایمنی می باشد. محققان با پیگیری بیشتر در صددند بتوانند ترکیبات دارویی متعددی را که به TSPO میتوکندریایی و دیگر مولکول ها ی مرتبط بیازمایند.
