ژانویه 30، 2013 - محققان از موسسه تحقیقات Benaroya در ویرجینیا (BRI) دریافته اند که پروتئین در مسیر سیگنالینگ IL-6 ممکن است به عنوان نشانه گرهای زیستی جدید بیماری ام اس (MS) برای ارزیابی فعالیت بیماری و به عنوان یک هدف جدید برای درمان های قوی تر باشد. این تحقیق مورد بررسی قرار گرفت تا ارزیابی کند چگونه اجزای دخیل در پاسخ ایمنی بین بیمار MS و نمونه شاهد متفاوت است،




تحت شرایط عادی، سلول های T موثر، ما را از عفونت ها و سرطان محافظت می کنند و کار سلول های T تنظیمی برای جلوگیری از حمله سلول های T موثر به بافت های سالم و در نتیجه جلوگیری از بیماریهای خودایمنی مانند MS است. MS زمانی اتفاق می افتد که سلول های T موثر سیستم ایمنی بدن به اشتباه به میلین، که سیستم عصبی مرکزی را احاطه و از آن محافظت می کنند حمله کند. هنگامی میلین آسیب دیده است، تکانه های عصبی با سرعت و یا موثر منتقل نمی شود و منجر به علائمی مانند بی حسی، ضعف، مشکلات بینایی، اختلال شناختی و یا خستگی، در میان بیماران می شود.

گروه باکنر نشان داد که سلول های T موثر بیماران مبتلا به ام اس عود کننده و فروکش کننده با بیماری فعال قادر به جلوگیری از سرکوب سلول های T تنظیمی بودند، در حالی که کسانی که از بیماران مبتلا به MS خفیف یا به خوبی کنترل شده بودند مقاومتی در برابر سرکوب توسط سلولهای تنظیمی نشان نمی دهند. این نتایج نشان می دهد که وجود یا عدم وجود مقاومت سلول های T موثر به سلول های T تنظیمی می تواند به بیماران و پزشکان اطلاعات ارزشمندی را در مورد سطح فعالیت بیماری و پتانسیل برای پیشرفت بیماری یک فرد بدهد. محققان همچنین کشف کردند که مقاومت در برابر سرکوب سلول های T در بیماران مبتلا به RRMS با افزایش حساسیت به IL-6، یک پروتئین که سیستم ایمنی بدن آن را تولید می کند و نشان داده شده است که در مقاومت به سرکوب سلول های T موثر شرکت میکند، افزایش می یابد.

گروه باکنر نشان داد که بیمارانی که مقاومت سلول T به سرکوب را به نمایش می گذارند نیز حساسیت بیشتری نسبت به IL-6 دارند. علاوه بر این، هنگامی که سیگنال های تولید شده توسط IL-6 در این سلول های T مسدود شد، مقاومت در برابر سرکوب، معکوس شد و این نشان می دهد که درمان با هدف قرار دادن مسیر IL-6 به طور بالقوه برای تنظیم مقاومت در برابر سلول های T به سرکوب می تواند مورد استفاده قرار گیرد.

دکتر باکنر گفت: "این یافته ها یک قدم هیجان انگیز به سوی درک بهتر این موضوع است که چرا MS رخ می دهد و کمک می کنند تا ما ارزیابی بهتری از درجه فعالیت بیماری در بیماران مبتلا به MS داشته باشیم و به در نظر گرفتن رویکردهای جدید برای درمان MS کمک می کند:"

"درمانی که مسیر IL-6 را هدف قرار می دهد در حال حاضر در دسترس برای درمان بیماری های دیگر خود ایمنی است و در در این تحقیق در MS مورد آزمایش قرار گرفته است."

جهت تحقیقات آینده به سمت بررسی نقش مقاومت سلول T به سرکوب و سیگنالینگ IL-6 در شروع MS و اینکه آیا سیگنالینگ IL-6 می تواند به عنوان نشانگرهای زیستی برای پیش بینی شدت بیماری در زمان تشخیص و یا پیش بینی عود و پیشرفت بیماری مورد استفاده قرار گیرد می باشد .
