سلول های چرب ( گلیال) به نورونهای عصبی مواد غنی از انرژی متابولیک عرضه می کنند.

کلاوس آرمین و گروه تحقیقاتی وی از موسسه ماکس پلانک در گوتینگن در حال حاضر مکانیسمی عملی کشف کرده اند که نشان می دهد سلول های چرب در مغز می تواند آکسون های مرتبط با خود را حمایت کرده و آنها را در دراز مدت زنده نگه دارد.

الیگودندروسیتها گروهی از سلول های چرب بسیار تخصصی در سیستم عصبی مرکزی هستند. آنها مسئول تشکیل میلین هستند که مانند غلاف اعصاب را میپوشاند و سرعت انتقال پیامها را افزایش می دهد و همچنین مصرف انرژی را کاهش می دهند.

جالب توجه است، عملکرد الیگودندروسیتها به مراتب فراتر از صرفا تولید میلین است. کلاوس آرمین و تیمش در موسسه ماکس پلانک در گوتینگن سالها پیش نشان داده بودند که سلولهای چرب سالم برای عملکرد بلند مدت و بقای خود آکسون، صرف نظر از میلین سازی ضروری می باشند. آنها می گویند: "راهی که در آن الیگودندروسیتها از آکسون های مرتبط با خود حمایت می کردند برای ما تا به حال روشن نبود". در یک مطالعه جدید، محققان توانستند نشان دهند که سلول های چرب، در کنار سایر وظایف، در دوباره پر از انرژی کردن رشته های عصبی نقش دارند. "آنها می تواند به عنوان پمپ بنزین در بزرگراه ها ی آکسون عمل کنند.

اما چگونه کار سوختگیری انرژی انجام می شود؟ آیا یک اتصال متابولیک بین الیگودندروسیتها و آکسون ها وجود دارد؟ برای پیدا کردن این مسئله، از موش های اصلاح شده ژنتیکی استفاده کردند که عملکرد میتوکندری در الیگودندروسیتهای آنها از طریق غیر فعال کردن ژن Cox10 مختل شده بود. این کار در مراحل نهایی تجزیه قند در میتوکندری که در آن انرژی ذخیره شده است و زنجیره تنفسی نامیده می شود تاثیر می گذارد .اگر ارتباط موجود در این زنجیره از دست برود، سلول های چرب به تدریج ظرفیت انرژی برای تنفس سلولی در میتوکندری خود را از دست می دهند. "بدون تنفس مستقل، سلول های چرب دستکاری شده سیستم عصبی می میرند مگر اینکه از انرژی محدودی که در اثر فرایند گلیکولیز ( شکستن گلوکز ) حاصل می شود استفاده کنند.

و این دقیقا چیزی است که دانشمندان در موش مشاهده کردند: میلین در موشها در ابتدا به صورت طبیعی شکل گرفت. از دست دادن زنجیره تنفسی میتوکندری، که در این نقطه آغاز شده، به نظر نمی رسید بر سلول های چرب در سیستم عصبی مرکزی تاثیر گذارند. حتی یک سال بعد، هیچ تغییری در بازسازی سلول های عصبی در مغز مشاهده نمی شود و بازسازی صورت می پذیرد. دانشمندان فرض می کنند که در اوایل تخریب - مرحله نیازمند به انرژی حداکثر - الیگودندروسیتها هنوز به میتوکندری های سالم باقیمانده تکیه می کنند. همه الیگودندروسیتها بالغ تر بعدا به نظر می رسد تنفس میتوکندری را کاهش می دهند و برای تنظیم آن تولید انرژی از طریق گلیکولیز افزایش می یابد. این عمل گلیکولیز در سلول های سالم چرب ، در هنگام سنتز میلین که فعالیت متابولیکی بالا است یک مزیت بزرگ می باشد وعلاوه بر این، اسید لاکتیک حاصل در الیگودندروسیتها به آکسون ها می روند تا برای تولید انرژی با کمک میتوکندری خود آکسون استفاده شود.
احتمالا از دست دادن کامل زنجیره تنفسی در الیگودندروسیتها یک گام تکاملی را در آنها افزایش می دهد". بنابراین از دست دادن میتوکندری در سلولهای چرب باعث وخامت عرضه انرژی به آکسون نمی شود، اما برعکس، به عرضه بیش از حد از اسید لاکتیک منجر می شود. عصب آسیب دیده بدون مشکل اسید لاکتیک را از الیگودندروسیتها دریافت می کند. پروتئینهای حمل ، انتقال سریع اسید لاکتیک در بین الیگودندروسیتها و آکسون را انجام می دهند.

این یافته ها درک جدیدی از نقش الیگودندروسیتها را فراهم می کند: علاوه بر اهمیت شناخته شده قبلی برای میلین سازی، آنها می توانند به طور مستقیم مواد گلوکزی را به آکسونها ارائه کنند که می تواند به عنوان سوخت با کمک میتوکندری آکسون در دوره هایی که شدت فعالیت ها بالا است استفاده شود. این مسئله اکنون می تواند توضیح دهد، چرا در بسیاری از بیماریهای میلین، به عنوان مثال ام اس، آکسون آسیب دیده اغلب دچار آسیب غیر قابل برگشت می شود.
