کنترل واكنش هاي حساسيتي به بادام زميني با فریب سیستم ایمنی

ساینس دیلی 11 اکتبر 2011 بر اساس یک مطالعه بالینی جدید از پزشکی نورث وسترن محققان توانسته اند با فريب دادن سيستم ایمني بدن موش ها (به سيستم ايمني القا مي شود پروتئين هاي بادام زميني تهديدي محسوب نمي شوند) واکنش های آلرژیک تهدید کننده زندگی بادام زمینی در موش ها را كنترل كنند
اين رویکرد شامل متصل كردن پروتئين بادام زميني به سلول هاي خوني است و سپس دوباره آن را به سيستم بدن وارد مي كند.
پل برایس ، دستیار پروفسور طب دربخش آلرژی ایمونولوژی دانشکده پزشکی فینبرگ در دانشگاه نورث وسترن گفت: ما فکر می کنیم که یک راه ایمن و سریع برای خاموش کردن واکنش های آلرژیک به حساسیت های غذایی ، یافته ایم.
برایس و استفان میلر ، استاد میکروبیولوژی و ایمونولوژی در فینبرگ ، نویسندگان اصلی مقاله منتشر شده در مجله ایمونولوژی هستند.
این اولین بار است که این روش برای ایجاد تحمل در سیستم ایمنی بدن در بیماری های آلرژیک استفاده می شود.
این رویکرد دارای سود ثانوی است وآن افزايش تعداد سلول هاي T است كه نوعي سلول هاي ايمني هستند و در شناخت پروتئين بادام زميني به عنوان پروتئين عادی نقش مهمي دارند.

برایس گفت : "سلول های T اشکال مختلفی دارند. اين روش سلول خطرناك Th2 T كه باعث ايجاد آلرژی و گسترش واكنش آن در بدن است را به نوعي ازميدان خارج می کند. تصور می شود پس از درمان مي توان بادام زميني را به راحتي مصرف كرد زيرا تعادل لازم در سيستم ايمني بدن به وجود آمده است.
آلرژی بادام زمینی اغلب باعث واکنش های آلرژیک تهدید کننده حیات ، به نام آنافیلاکسی می شود . بر اساس تحقيقات موسسه ملي سلامت، سالانه در امريكا 15 تا 30 هزار نمونه واكنش هاي مرتبط با آنافیلاکسی و 100 تا 200 نمونه مرگ مرتبط به اين حساسيت ها ثبت مي شود. هیچ درمان امن برای محافظت ازافراد از یک واکنش آلرژیک شدید به غذا وجود ندارد.
هنگامی که یک فرد حساسیتی بادام زمینی می خورد ، پروتئین ها از طریق روده جذب می شود و می تواند تهدید کننده حیات باشد . این شامل تنگ شدن راه های هوایی ، فشار خون پایین و / یا شوک می شود و می تواند منجر به از دست دادن آگاهی و مرگ گردد


محققان اين روش را روي موش هاي نمونه يي انجام دادند كه داراي حساسيت كشنده نسبت به بادام زميني بودند. (تیم نورث وسترن )آنها پروتئین های بادام زمینی متصل بر روی سلول های سفید خون به نام لکوسیت را به موشها تزریق کردند .
پس از دو مرتبه درمان، به موش ها عصاره بادام زميني داده شد و آنها هيچ واكنش آلرژيكي نسبت به آن نشان ندادند زيرا سيستم ايمني بدن شان دیگر آن پروتئين را ايمن شناخته بود
برایس گفت : سيستم ايمني بدن پروتئين بادام زميني را نوعي ماده كاملا عادی تشخيص مي دهد زيرا اين ماده قبلابه سلول هاي خوني وارد شده است. بدون اين روش درماني، موش ها پس از خوردن عصاره بادام زميني دچار شوك آنافیلاکتیک مي شدند.
برايس عقیده دارد می توان بيش از يك نوع پروتئين را به سطح سلول هاي خوني متصل كرد تا به اين ترتيب آلرژی های غذایی مختلف در یک زمان درمان شوند.
در بخش دوم تحقيقات، محققان همان روش را براي نوعي ديگري از آلرژی غذايي آزمايش كردند (پروتئين تخم مرغ) كه واكنش سيستم ايمني بدن به آن تا حدودي شبيه حمله آسمي به شش هاست. دوباره پس از آنكه پروتئين ها به سلول هاي T متصل شده و دوباره به بدن موش ها تزريق شد، هيچ گونه اثري از حملات آسمي در آنها ديده نشد.
میلر ، جودی گوگن هیم استاد تحقیقات در دانشکده فینبرگ گفت :" راه های جدید بسیار هیجان انگیز است وما می توانیم بیماری های خاص آلرژی را حل كرده و تنظیم کنیم در نهایت ممکن است در یک محیط بالینی برای بیماران استفاده نماییم.
روشي كه محققان دانشگاه پزشكي نورت وسترين فينبرگ ارايه كرده اند مي تواند تمام اين مشكلات را حل كرده و حتي سبب مي شود افراد مبتلابه حساسيت (هر نوع حساسيت غذايي) بتوانند از غذايي كه مدت هاي مديد، از آن محروم بوده اند لذت ببرند.
میلر همچنین همین رویکرد را در بیماریهای خودایمنی بکار برده است. تحقیقات قبلی منتشر شده او به روش های مشابه برای متوقف کردن پیشرفت بیماری ms ودیابت نوع 1 و بیماری های خود ایمنی در مدلهای حیوانی نتایج خوبی نشان داده است. این رویکرد در حال حاضر در بیماران مبتلا به مولتیپل اسکلروز یس در فاز I / IIA کارآزمایی بالینی تست شده است.
میلر همچنین برای بیماری های خود ایمنی و بیماری های آلرژیک تنفسی ، در حال کار با میکروذرات به جای سلول های سفید برای القاء تحمل است ، زیرا میکروذرات آسان ترواستاندارد تر برای ساخت هستند

فرق سلول Th2 T با سلول Th1 T را کسی میتونه توضیح بده؟

Th1 از طریق فعال کردن ایمنی سلولار عمل میکنه و به عمل ماکروفاژها کمک میکنه ولی Th2 به تولید انتی بادی کمک میکنه.

من وقتی بادام زمینی میخورم از نظر حرکتی مشکل پیدا میکنم ، این یعنی همین حساسیت ؟

می تونین برین آزمایش بدین تا ببینین نسبت به چی حساسیت دارین.من 11 سال قبل رفته بودم و آزمایش دادم.اون موقع نسبت به هیچ جیز حساسیت نداشتم.

اقای بهرنگ خان چرا نوشتین باز هم اوونکس یعنی چی ؟ یعنی قطع کرده بودین و باز دوباره شروع کردید یا داروتون عوض شده بوده و دوباره به اوونکس برگشتید ؟confused2
