اثرداروی سولیفناسین درمیلین سازی و درمان ام اس - sia - 2015/03/11

یک داروی موجود می تواند به درمان MS  و سایر بیماریهای عصبی کمک کند.

آسیب به میلین که یک عایق چربی است که موجب ارتباط بین سلول های عصبی می شود، از ویژگی های MS و برخی آسیب های عصبی دیگر است. این آسیب به طور یکدفعه ایجاد نمی شود، در واقع معمولا دوره ای از میلین سازی مجدد اتفاق می افتد که این توانایی بازسازی و ترمیمی در برخی بیماران با افزایش سن و برخی بیماری ها از بین می رود.

به گزارش بنیان به نقل ازnews-medical.net، اینک در مطالعه ای محققین در بوفالو با استفاده از یک دارو که از قبل در بازار وجود دارد توانسته اند این توانایی میلین سازی مجدد را حفظ کنند.

آن ها در این مطالعه از دارویی که برای پیشرفت سلول درمانی استفاده می شود برای بیماری های میلینی استفاده کردند و در یافتند که می توانند با هدف قرار دادن سلول های پیش ساز اولیگودندروسیتی با داروی سولیفناسین(solifenacin) که یک داروی ضد موسکارینی است موجب افزایش میلین سازی شوند.

فرضیه این محققین این بود که در MS، به نظر می رسد سلول های پیش ساز اولیگودندروسیت به نوعی گیر افتاده اند و نمی توانند بالغ شده و به اولیگودندروسیت های میلین ساز تبدیل شوند.

آن ها در این مطالعه مسیر سیگنالینگ دخیل در این بلوغ را شناسایی کردند و سپس کاندیدای دارویی که موجب می شود که این تمایز سلول های پیش ساز به اولیگودندروسیت صورت گیرد را شناسایی کردند.
آن ها دریافتند که زمانی که گیرنده موسکارینی نوع 3 روی سلول های پیش ساز اولیگودندروسیتی فعال شد، تمایز به طور کامل متوقف شد. حال اگر ما چیزی داشته باشیم که بتواند به جای بلوک کردن موجب تحریک این فرایند شود، می توانیم تمایز را افزایش دهیم.

به این منظور آن ها داروی سولیفناسین را استفاده کردند که دارویی بود که مورد تایید FDA بود و در بازار نیز موجود بود. به منظور این که ببینند آیا دارو می تواند سنتز میلین را افزایش دهد، محققین سلول های پیش ساز اولیگودندروسیتی انسانی را به موشی که نمی توانست میلین بسازد پیوند کردند. نتیجه این امر افزایش تمایز و سنتز میلین از این سلول ها بود. اما آن ها احتیاج به یک تست عملکردی نیز داشتند تا بدانند که میلین حاصل از این سلول ها وظیفه خود را به درستی انجام می دهد. آن ها از تست های شنوایی ساقه مغز استفاده کردند که فعالیت موج های مغزی را ثبت می کند. نتایج نشان داد که پاسخ ها به این سیگنال های شنوایی در این جانوران که پیوند سلول های پیش ساز اولیگودندروسیتی را دریافت کرده و با سولیفناسین تیمار شده بودند بهبود یافته است.

بنابراین به نظر می رسد محققین راهی را برای بهبود میلین سازی انسانی یافته اند.
